In Vitro Cytotoxicity Activity of a Staphylococcus Haemolyticus Bacteriocin Against Prostate Cancer Cells

Abstract

Cancer is regarded as one of the most common and leading causes of death.Even while there are conventional therapies for cancer cells, they are not the best option because they run a chance of having a number of unfavorable side effects on the body. The aimed alternative An antimicrobial peptide created by bacteria called bacteriocin is one of the promising new therapeutic possibilities for cancer treatment, that doesn't have any additional adverse effects. Bacteriocins producing and optimization from S.haemolyticus isolated from semen fluid and urine included the were chosen randomly from Baghdad they were  males and femal  with 18 -45years were identifection to macroscopic confirmation identification  by VITEK2 system, The optimazation  for bacteriocin condition production in modification media with 1% glucose at 37°C for 72 hr.  with inoculum size (1.2 ˟ 109 CFU/ml) and pH 7 .In order to screen antibacterial activity of bacteriocin that produce against bacteria. The S.haemolyticus. Screening bacteriocin production from S.haemolyticus was showed that the Filter paper disc  (FPD) method. Bacteriocin wase  best  produce S.haemolyticus, bacterial activity of bacteriocin was at 70 and 40 AU/ml After partial purification of bacteriocin by ammonium sulfat  method  at saturation reached 70% with Mwt reached 29 kDa . The cells viability was decreased with increasing the concentration of Bacteriocin and the IC50 values of the HdFn cells (2.5 µg/mL).In conclusion, S. haemlyticus   bacteriocin could be a good antibacteral agent and acivity against other types of species isolation semen fluid and urine clinical specmen.
